She doesn't really have any set starting moves in combat afaik but she'd prob lead with giving herself a Power Up and then using her magic abilities like Sleep Time and Mute.

I think Tsunade's profile indicates that she's Island level to start with and gets far higher with Byakugo. Then again, Peach can still get higher because, surprise!
