Maybe it's something we all know unanimously, but I'm a little confused about it.

Afaik, destroying regular space-time (which is 3 spatial + 1 time) it's a requirement for Low 2-C. So, how about destroying 4 spatial dimensions but have nothing to do with temporal dimension? It's still Low 2-C or not.

If it has size equals to Universe in 3 dimensions, sure.
 
Then, which one is stronger between Low 2-C from 3 spatial + 1 time and Low 2-C from 4 spatial dimensions. Or it depend on other contextual factors?

Size matters. For any finite 4 dimension or size less than standard Universal model will be Tiered less than standard Universal model.
 
However, there is a situation like this, someone who manipulates only a 4-D spatial dimension gains only 4-D spatial dimension manipulation, while someone who can control space and time has space and time manipulation, and this puts him one step ahead, I think.
